Macoupin County is a county located in the southwestern portion of the U.S. state of Illinois. The population was 49,019 at the 2000 census, and the population was 48,235 in the 2007 official US Census estimate. The county seat is Carlinville. Macoupin County is an outlying county of the Metro-East region of the Greater St. Louis metropolitan area.

The primary industry is agriculture, consisting of crops of corn (maize), soybeans, and some wheat.
