Lafayette County is a county located in the U.S. state of Missouri. As of 2010, the population was 33,381. Its county seat is Lexington . Lafayette is part of the Kansas City Metropolitan Area.

The county was organized 16 November 1820 (effective 1 January 1821) from Cooper County and was originally named, "Lillard," for James Lillard of Tennessee. On 16 February 1825, the name was change to "Lafayette," for the Marquis de La Fayette.
